 Acquired aplastic anemia (AA), a bone marrow failure characterized by pancytopenia and bone marrow hypoplasia, is an autoimmune disease caused by cytotoxic T cell (CTL) attack against autologous hematopoietic stem cells (HSCs).

Autoantigens capable of eliciting the CTL against HSCs in AA have been extensively studied by many researchers over the years, but none of the studies have been successful so far.

The failure in identifying autoantigens of AA is mainly due to a lack of HSCs that are necessary to establish CTLs from patients’ T cells specific to autoantigens presented by HSCs in vitro.

I plan to produce HSCs from induced-pluripotent stem (iPS) cells that are induced from AA patients’ B lymphocytes.

The sufficient number of HSCs from iPS cells should allow us to establish CTL clones specific to autologous HSCs and also to identify target autoantigens of the CTL clones using the expression cloning method.
